You should call and find out the name of the person to whom the letter should be addressed. This shows that you have done some research on the position and on the hiring process, and it will help ensure that your application package ends up on the right desk by labelling the envelope "Attention: So-and-so."

Even if they can't give you a name, they could give you the title of the person who will be hiring your position. It may even be a hiring committee that takes care of it, in which case you could say "Dear Selection Committee" (which is common in Universities). Otherwise, you could say "Dear Mr. Jones" or "Dear Sir or Madam." Remember to use a colon instead of a comma to be more formal.
