A cover letter gives you an opportunity to show why you are the best person for the job. It should be in a standard business format and contain four parts.
Header: Your address, date, the recipient's name and address.
Introduction: Briefly state the specific position applying for.
Body: Highlight something from your resume that would indicate your interest and value to the employer. Any skills, qualifications or past experience could be noted.
Closing: This concludes or sums up the letter, such as I look forward to hearing from you. Then the closing (Sincerely) followed by your name.
Keep the letter simple, no one is going to read through hundreds of these. Give the employer a reason to read your resume and consider you.

Dear Mr. Smith,

In this paragraph, indicate why you would be a great bar staff person or your past experience. Talk about your people skills, meeting people, taking initiative, flexibility, reliability, communication skills, etc. Pull out the main highlight of your resume.

In the closing paragraph, something like: I greatly appreciate your consideration of my candidacy for this position and look forward to meeting you. I will be able to start work immediately when the hotel opens and can be reached at 999-999-9999.

I believe that sentence refers more to working conditions. They mean that they don't want to have to change the layout of the office for you to work there, not that they would not be willing to train you. I would emphasis your strengths in the letter. I would also mention your desire to learn and grow in the business/company. You can bring the rest up at the interview. Don't lie about what you can do, but don't emphasize what you can't.

Your cover letter should show confidence that you are able to do the job and would be a good fit for it. Emphasize the skills you do have and how you see yourself as a great candidate for it.

How to handle the areas in which you fall short depends on how far off you are and how glaring the shortcomings are. If they ask for 2 years of experience and you have 6 months, you may have to deal with that by pointing out other experience or how well you were trained during those 6 months. If they want a BA but you have just a HS diploma, you'll need to explain how you've learned things on your own, on the job, etc.

If your resume is a poor fit to the requirements, they'll toss you out quickly. The worse the fit, the more you'll need to make a good impression in your letter.
