I am a professional resume writer (which also means cover and thank you letters), so please listen.In your letter, be cordial, yet still professional. You are not her friend, or her colleague. You are looking for a job.
Make sure you thank her for her time, and consideration. Stress how interested you are in the position.
Hopefully during the interview you took notes (or have a superb memory). Recap the points in your skills that make you the best candidate. If she voiced any concerns about your ability to do the job, address those as well- but make sure you give supporting evidence as to why those are not things to be concerned about. for example- if a concern was inexperience- you can refute that by emphasizing your willingness and quickness to learn from other staff members.
Run spell check, but also have a friend (who is detail oriented and would notice mistakes) read it. Go have a snack or run an errand and proof read it again before sending it.

These are competitive programs. Your GRE score will help tremendously. Your qualifications are all in order, so I wouldn't worry about that.The big factors in your case will be "fit" and the competition.
You've indicated that there are indeed faculty in these programs with specializations in your area of study, and that is critical when the faculty examine your application for "fit." The level of competition is unpredictable from year to year, however.
Without seeing your entire application, I can't really weigh your chances, but it seems like you've got a good shot, and should be admitted to more than one of your schools.
If possible, contact the DGS at each program to arrange a visit. Speaking in person to some of the faculty could be helpful in terms of admissions. But don't go directly to faculty. Tell the DGS with whom you'd like to meet, and see if it is possible. Some programs can even help defray some of the cost of your travel, and/or put you up on campus. (My program does.)
Just remember when you do receive your acceptances to weigh BOTH the level of support extended to you AND the reputation of the program.

Then the company and address to whom you are writing (laid out as it would be on the envelope)
Then put 'For the attention of (put a name in here)
Start Dear Sirs
End Yours faithfully
with your name below signature.
Look at real business correspondence to see what it should look like.

These references are meant to convey one's character...just as simple as that. When you think of the person, what qualities come to mind? What are some experiences that you've had with them that have been positive? Think of those answers and tie them into how the person would perform on the job.
