If it has been a while since you prepared a resume and job-hunted, you really should invest in a professionally prepared resume, etc. The market has changed so much that many of the library books on resume preparation, cover letters, etc, are out of date.

As for the cover letter, no more than 3 paragraphs, succinct, personalized for each individual and company you send it to. There's more, too much more to put in a post. Get a pro.

Don't sweat the small stuff, if you have good qualifications it will be passed on, generally the human resource director or her secretary will receive applications...if you addressed anyone else only their secretary will open it and forward it to human resources. Your cover letter transfers your skills to the requirements of that employer. If they want someone with 2 years office experience your cover letter must reflect those years with your accomplishments. An employer really wants to know what you can do for them (employers only hire people so they can make the employer money; positively affect the bottom line).so no one cares why you want the job. They care about what you can DO for them.

A cover letter to accompany a CV should be very brief but have enough information that the reader understands what you are sending him.

The tip I have found helpful is to remember you are not repeating your CV. Loads of people make the mistake of just blethering out their CV again but the point of a covering letter is to highlight out your best points - achievments, skills, what you could bring to the company, etc. Keep it short and sweet and no more than a couple of paragraphs.
